Comparison of factor deficient plasmas for measuring Factor VIII and IX
نویسندگان
چکیده
Introduction Factor dosages are part of the diagnostic work up in patients with bleeding tendency (factor (F) VIII and FIX) or in thrombophilia screening (FVIII). The correct diagnosis of FVIII and FIX deficiency and the assessment of severity of the disease are essential for a patient-tailored treatment strategy. Persistently high levels of FVIII are regarded as a thrombophilia risk marker. FVIII can be measured using the one-stage clotting assay, two-stage clotting assay and the chromogenic (amidolytic) assay. The one-stage method suffers from diversity in the reagents and assay conditions used: difference in activated partial thromboplastin time (aPTT) reagents, deficient plasmas, reference plasmas, and instruments.

An alternative extrinsic pathway of human blood coagulation.
To study the interrelationships of the major human coagulation pathways, factor X activation in normal and various deficient human plasmas was evaluated when clotting was triggered by dilute rabbit or human thromboplastin. Various dilutions of thromboplastin were added to plasma samples containing 3H-labeled factor X, and the time course of factor X activation was determined. At a 1/250 dilutio...
متن کاملImmunologic identification of tissue factor (thromboplastin) synthesized by cultured fibroblasts.
Immunologic methods were employed in an attempt to identify a potent procoagulant present in homogenates of human skin fibroblasts cultured in vitro. The activity of this procoagulant was restricted to the early stages of coagulation and was heretofore considered to be due to tissue factor (tissue thromboplastin, factor Ill) either alone or in combination with one or more of the first-stage coa...

Background: During extrinsic coagulation pathway, a complex is developed between factor VII, calcium and tissue factor (a cell membrane lipoprotein that is exposed after cell injury). Factor VII needs calcium and vitamin K for its biologic activation. Coronary artery disease can be induced by increased level and activity of the coagulation factors VII, VIII and IX. In postmenopausal period, est...
متن کاملCoagulation-dependent inhibition of fibrinolysis: role of carboxypeptidase-U and the premature lysis of clots from hemophilic plasma.
Coagulation is initiated by the binding of factor VIIa to tissue factor, with resultant limited factor IX and X activation and thrombin production. Owing to the feedback inhibition of the factor VIIa/tissue factor complex by tissue factor pathway inhibitor (TFPI), additional factor X activation and thrombin generation must proceed through a pathway involving factors VIII, IX, and XI. Experiment...
متن کاملThe Survey of Effective Agents on Factor VIII and IX Inhibitors in Patients with Hemophilia A and B in Kermanshah Province
Background: Hemophilia is the most frequent severe hereditary hemorrhagic disease due to deficiency of coagulation factors VIII (Hemophilia A) or IX (Hemophilia B) in plasma. We aimed to identify patients with hemophilia in Kermanshah, Iran and assess the incidence of inhibitors in this population and its associated factors. Methods: This study was conducted on patients with hemophilia...
